Description:
Direct cloning of PCR products can be achieved using GenoMechanix’s pMAK vector that contains single T 5’-overhangs.
Taq amplified DNA contains
single A 3’-overhangs which are complimentary to the T overhangs in the
pMAK vector. DNA amplified by
other thermostable polymerases that leave blunt ends can be cloned by simply
incubating the blunt end DNA with GenoMechanix’s Taq
polymerase to add 3’-A overhangs.
The kit comes with all the necessary components to perform the PCR
reaction. The user supplies the specific primers and DNA template.
T4 DNA Ligase is included to perform the ligation step.
Competent cells are also included for transforming with the pMAK/PCR
product ligation.
