PRIMERS FOR MAMMALIAN CLONING/SEQUENCING/EXPRESSION ANALYSES
The primers in this section
have been designed for the viral vectors commonly used for cloning and
expression of therapeutic
proteins in mammalian cell culture and also in gene therapy research.
Recommended regulatory guidelines for the characterization of cell
lines used to express proteins frequently involves an examination of the
potential heterogeneity of the mRNA transcripts in the master cell bank and
the manufacturer’s working cell bank.
Amplification and sequencing data on mRNA from cells transformed or
infected with the respective viral vectors can be obtained using the primer
sets listed in this section. Our
primer pairs may be used to amplify and sequence mRNA during the production
of a virus preparation used in gene therapy.
These analyses may be required for initial characterization of master
and working cell/virus banks and for ongoing stability studies. Characterization (amplification, sequencing) of the viruses
and mRNA species present in tissues derived from animals used in efficacy
and toxicity testing may also be performed using the relevant primer sets
listed below.
