RANDOM PRIMERS
Random
primers are useful in the identification of variation among
species through RAPD (Random Amplified Polymorphic DNA)
analysis. Random
primers are used to amplify genomic regions for analysis of
polymorphisms. The
protocol used involves denaturation at 95oC for 5 min
followed by cycles at 95oC for 30 sec, 30oC
for 2 min and 72oC for 1min.
This should be carried out for 33 cycles with the
annealing temperature changed to 40oC.
The final cycle the extension step should be for 5 min (Hamid,
et. al., 1999, Rev. Inst. Med.
Trop. S. Paulo; Vol. 41, no. 5).
Longer random primers typically detect fewer
polymorphisms and are best suited for more distantly related
species.
